I guess you don't have to know the manga to figure this one out but the anime pretty much gives us the whole picture. Most of it at least. I wanted to figure out just how closely related the souma's are and it could maybe have a clue on how the curse is passed down. ^^
First, we know that the following people are siblings. This might contain spoilers but if you want to be spoiled, that's the only time I'll give you the info. ^^
Yuki(mouse) and Ayame(snake) are brothers
Rin(horse) and Hiro(goat) are siblings
Shigure is Yuki and Ayame's Cousin. Yuki introduced Shigure as his cousin to Tohru in the first episode.
Hatori is a "relative" which means he might be a 3rd, 4th or even 5th cousin. Shigure mentioned this when he came to pick up Tohru at Hatori's house. This was Tohru's first visit at the main house.
Those are the only close relatives that I can remember right now and everyone else was mentioned as a "relative." Another point I want to make clear that it is ILLEGAL to marry your brother, sister or FIRST cousin in Japan. Any romantic relations that might have happened (hatori x hana; kisa x hiro) are assumed that those people are not closely related.
As for the curse, it seems like it's passed down by the males in the souma family.
Females seem to be a rarity in the Sohma family. There are only 3 female members of the jyuunishi.
In Momiji's case, his mom was German and his father was definitely a Sohma. His father passed down the gene that might be causing the curse to momiji.
These are the only two reasons that I can think of. o_O I don't know how this might be related but you also have to think about Kyo's grandfather. Remember, anyone who is cursed changes to an animal when hugged by an opposite sex. Since he was pretty much locked up in a cage all his life, there's a huge possibility that he was married to another jyuunishi at the time.
